Question

Lab Report 2: PCR and qPCR

REMINDER: We do not want either a repeat of the Lab Manual or a repeat of everything

you recorded in your Lab book. (Marks will be deducted for failing to adhere to this.)

1. Briefly describe the steps involved in a Polymerase Chain Reaction.

2. Attach a properly labelled photograph of amplicons from PCR. Estimate the sizes

of any "bands" or size range of any "smears" that can be discerned from the

Ladder.

3. Present a table of your NanoDrop quantification of "purified PCR products".

Comment on the yield and the purity of samples. Be sure to address any

anomalies.

4. Briefly describe the "Types" of qPCR used to quantify gene expression. Comment

on the type of qPCR you have used to quantify the expression of your gene of

interest.

5. Provide a table of your qPCR data analysis and present the results in a bar diagram

with proper labeling.

6. Summarize the results from your qPCR experiment. Comment on whether these

results reinforce or contradict your initial hypothesis on the expression of your gene

of interest in Ras-transformed cells compared to the control cells?

Additional Resources if you need them:

• The sizes of the DNA fragments in the DNA Ladders are in Appendix #2.
